Also we not sure we have power to the 24vac, cause we do have power to the AC unit outside, and we got power to the furnace cause we testing with voltage meter and we got sparks by going too deep across metal breakers, and still got power now!Plenty to cause yourself serious injury. The installation instructions were generally easy to follow, but I also found additional information on the Honeywell website explaining what each terminal actually was. It would have been useful for Honeywell to print this in the instructions. Summarized: Rh: Heating power supply Rc: Cooling power supply W: Heating signal Y: Cooling signal G: Fan The two heating systems in my home are both ONLY heat, and have a two-wire connection. The wiring looked about a hundred years old, but still worked perfectly once I connected the wires to the Rh and W terminals. Interesting to note that in both installations it did not seem to make a difference which of the two wires went to what terminal, so long as it was Rh and W. However in a more complicated system that includes more wires for cooling or a fan, I'd use the included labels to mark each wire and be certain it was connected properly. I don't have any dollar amount savings to offer, but I can say that the temperature was MUCH more consistent in my apartment than it had been with the old round-style mercury switch which was prone to large temperature swings. There is no question in my mind that less fuel was used, and it's very likely that the two thermostats I purchased have already paid for themselves one month later. I love it! I used to worry that my HVAC equipment could be damaged if the mercury ball bounced on closing. No more problem! Setting the temperature used to be vague. No more problem! I used to get chilly waiting for the heat to come on and hot waiting for it to shut off. No more problem! In the cooling season, the mercury thermostat would call for progressively lower temperatures after it was turned on. No more problem! Restarting a compressor too soon could damage it. No more worry! Connecting the wires was unexpectedly troublesome. I straightened the ends with my fingers, stuck them in the terminals, and tightened. I couldn't get them really secure until I used pliers to get the ends really straight.
Also, the wiring with the electrical tape covering could also be the issue if the wires are broken internally (most t-stat cable is solid wire), it will intermittently short on and off whatever that wire is controlling. Not for any actual exchange or installation of a new one. Depending on what thermostat I end up getting I will probably call one to make sure everything is done correctly. Just not that one. The job would have been easier if the instructions had specified the best screwdriver and mentioned getting the wire ends really straight. If you shut off the thermostat while you're away or to remove it, it's important to see if it's switched to Cool, Off, or Heat. The print was so small and pale that I had to turn on a light and get my glasses. I cut out the little square of paper, applied it to the thermostat with clear tape, and blackened the switch with a magic marker. Now I can see how it's set from across the room in low light. Update: Early in the fourth heating season, I saw the furnace cut off while the thermostat was still calling for heat. I couldn't find a reason, and I couldn't repeat it. The chimney blower comes on for perhaps 40 seconds before the burner lights. Late in the season, I heard the blower cut off twice during those 40 seconds. Suddenly it doesn't work and we have to twist the cover again to make it work. This has been going on for about 2 years. Fc retention was dominated by calcium affinity, while retention of Fab was dominated by cation exchange. The retention curve for intact IgG incorporated the distinctive elements of its fragments but stronger retention than that predicted by their addition to one another. Aggregate retention paralleled the curve for non-aggregated antibody, with stronger retention by both binding mechanisms. Experimental data revealed evidence of charge repulsion between IgG carboxyls and HA phosphate at low conductivity values. Electrostatic repulsion of amino residues and attraction of carboxyls by HA calcium appeared to be blocked by strong complexation of calcium with mobile phase phosphate. In addition to the economic value of these applications, the well-characterized structures of IgG and related entities make them suitable probes to investigate the surface reactivity of HA. The above studies, reporting on about 20 IgG monoclonal antibodies, have demonstrated that IgG is retained dominantly by a combination of calcium metal affinity and phosphoryl cation exchange, regardless of subclass, light chain type, or p I. These studies have also contributed important refinements to our understanding of IgG interactions with HA. A number of interactions predicted by studies on peptide and protein models remain uncharacterized. HA phosphates may repel protein carboxyls, in addition to their documented ability to bind amino groups. Dissecting the individual influence of these potential interactions is complicated by the arrangement of calcium and phosphate on the crystal surface. Cooperativity is described as positive in cases where multimodal interactions produce progressively stronger binding and negative where interactions depress net retention. It is important to note that the conditions under which many of the above experiments were conducted may damage HA. Corresponding sodium chloride concentrations are presented in Table 1. Retentions of lysozyme and BSA are plotted in the same figure to represent the behavior of cation exchange-dominated and calcium affinity-dominated proteins. It failed to elute in sodium chloride gradients at phosphate concentrations up to 40 mM phosphate but failed to bind at 60 mM phosphate ( Fig. 2 ). Its steep response curve resembles the early part of the IgG curve. The 40 mM horizontal offset in phosphate concentration between the curves indicates that IgG has much lower calcium affinity than BSA. This would also be understood to limit the number of polycarboxyl domains configured appropriately to support coordination with HA calcium. It elutes in a sodium chloride gradient even in the absence of phosphate, and exhibits a shallow linear decline in retention as phosphate is added to the elution buffers. It resembles the latter portion of the IgG retention curve, except that retention of IgG is much stronger. Rituxan’s p I is slightly lower than lysozyme. This suggests that its stronger cation exchange binding reflects a larger number of interactions, consistent with the more than tenfold greater mass of IgG, but it could also indicate a cooperative contribution by calcium affinity. Chloride acts primarily on ion exchange interactions; it has little effect on calcium affinity. The inherent limitation of these studies is that they have mostly been conducted for the purpose of aggregate removal, and the ranges of the key variables have been limited to practical operating ranges for HA. In the present study, we evaluate more extensive grids of phosphate gradients at different sodium chloride concentrations, and sodium chloride gradients at different phosphate concentrations in the hope of refining the model of IgG retention on HA. Immobilized papain and pepsin was obtained from Thermo Fisher Scientific, Rockford, IL, USA. A monoclonal antibody containing about 40% aggregates was a generous gift from Avid BioServices, Tustin, CA, USA. Briefly, the antibody and immobilized enzyme gel were equilibrated to the specified buffer. The immobilized enzyme was removed by centrifugation. The sample was eluted with a 20 column volume (CV) linear gradient to 20 mM Hepes, 1.0 M sodium chloride, pH 7.0. The column was cleaned with 500 mM sodium phosphate, pH 7.0. In the next experiment, the equilibration, wash, and elution buffers contained 5 mM sodium phosphate. In subsequent experiments, both buffers contained 10, 20, 40, 80, or 160 mM sodium phosphate. A corresponding series was conducted at pH 8.5, substituting 20 mM Tris for Hepes. Another series was developed by conducting phosphate gradients at different chloride concentrations. The column was equilibrated with 20 mM Hepes, pH 7.

The lysozyme curve in Fig. 3 ascends at a much shallower angle than BSA and rapidly converges with the gradient start line. That it ascends at all illustrates a positive contribution by calcium affinity. If binding were exclusively electrostatic, the curve would be horizontal. Calcium affinity is weak, however, as illustrated by non-retention at 160 mM sodium chloride. Convergence of the lysozyme curve with the line marking conductivity at gradient start signifies loss of electrostatic binding with increasing chloride concentration. This is understood to represent cation exchange between HA phosphate and lysozyme amino residues. The slope of the IgG curve is essentially parallel with lysozyme at 80 mM sodium chloride, possibly reflecting its similar p I. The IgG slope diverged from lysozyme at 160 mM sodium chloride, becoming more positive, conforming more closely to the line marking conductivity at gradient start, and thereby revealing its stronger calcium affinity. The BSA and IgG curves in Fig. 3 exhibit depressed retention in the absence of sodium chloride. A similar phenomenon was observed for lysozyme in the absence of phosphate ( Fig. 2 ). These features suggest the influence of interactions that weaken retention. Weaker retention absent sodium chloride was attributed to restricted contact of DNA phosphates with HA calcium due to electrostatic repulsion between HA phosphates and DNA phosphates. Sodium chloride was believed to suppress this effect, allowing DNA to participate in more coordination events with HA calcium.
Data points below the line of conductivity at gradient start indicate that the solute was unretained under the specified conditions. The most striking feature of the figure is that elution conductivities ascend with increasing sodium chloride concentration. At sodium chloride concentrations greater than 10 mM, the BSA curve ascends nearly parallel with the line created by the conductivity at gradient start for the various experiments. This demonstrates virtual immunity to sodium chloride, which in turn indicates the weak contribution of electrostatic interactions. The steeper slope of the Fc curve compared with IgG, and stronger binding than Fab in the absence of phosphate, suggest that Fc calcium affinity is intermediate between the two. Phosphoryl cation exchange for Fc was the weakest of all fragments: it failed to bind HA at 40 mM phosphate (5.9 mS). Data from phosphate gradients at different salt gradients support identical conclusions ( Fig. 5 ). Weaker Fc retention at all conductivities illustrates lower cation exchange binding, and the closer conformance of the slope with the line indicating conductivity at gradient start illustrates the relative dominance of calcium affinity. It makes sense that the cation exchange component of binding should be reduced since proteins generally become less electropositive with increasing pH. This would be expected to disproportionately reduce retention of solutes dominated by calcium affinity. Sodium chloride concentration and conductivity values are presented in Table 3. All examples suggest cooperativity, but most manifest too many confounding influences to express it with precision. Tetra-aggregates bound more strongly than IgG, by both calcium affinity and cation exchange, but not by a factor of 4 ( Fig. 7 ). This makes sense since associations among the individual IgG components must inevitably block some interactions with HA. This fits the definition for negative cooperativity. It is noted throughout the above discussion that various aspects of IgG and fragment retention are consistent with their p I s within an experimental series. This is not to suggest that p I is predictive of HA retention as a general matter. The greater the vertical offset between the elution conductivities for aggregated and non-aggregated antibody at a given phosphate concentration, the greater the separation. Figure 7 thus recommends that a sodium chloride gradient be applied at 5 mM phosphate. We evaluated aggregate separation in phosphate gradients at different salt concentrations but observed no separation under any conditions. All buffers can be made as dilutions of the 500 mM phosphate-cleaning buffer, and the elution conductivity of the antibody is usually low enough to support application to an ion exchanger with little more than moderate pH titration and dilution.
BSA is far less charged than DNA but still heavily carboxylated, suggesting that its retention might also be depressed to some degree by repulsion from HA phosphates. The parallel low conductivity response of Rituxan equally suggests the effects of charge repulsion, but this conclusion is challenged by its low carboxylation relative to BSA. This could be the explanation, nevertheless. If the number of IgG carboxyls not involved in calcium affinity interactions were roughly similar to BSA, negative charge repulsion at low conductivity should also be similar. Rituxan’s high p I raises the possibility that the lower retention absent NaCl might reflect charge repulsion between HA calcium and IgG amino residues, but this is challenged by the behavior of lysozyme. First, lysozyme’s retention in Fig. 3 does not exhibit the low conductivity depression manifested by IgG. Second, lysozyme’s behavior in Fig. 2 points to a specific mechanism by which all kinds of electrostatic interactions with HA calcium might be eliminated. We suggest that similarly strong pairing between HA calcium and mobile phase phosphate ions eliminates electrostatic interactions involving HA calcium. Figure 2 suggests that 5 mM phosphate is sufficient to suspend electrostatic repulsion of lysozyme. If sufficient for lysozyme, it should be sufficient for IgG. Lysozyme fails to show the effect in Fig. 3 because it elutes far above that threshold, at about 85 mM phosphate ( Table 2 ). Note that this hypothesis also predicts that HA calcium should be incapable of electrostatically binding protein carboxyl residues in the presence of greater than 5 mM phosphate. Thus, it appears, in the presence of phosphate, that HA calcium is able to participate solely in coordination interactions. Ion exchange chromatography results were consistent with the above data and interpretations. Rituxan failed to bind to the anion exchanger at pH 7, which would have been predicted by its alkaline p I.
It bound strongly to the cation exchanger at the same pH and eluted at 11.3 mS (135 mM sodium chloride). It bound to an HA column equilibrated at 10.4 mS (80 mM phosphate, Table 1 ), but failed to bind HA at 160 mM phosphate. This demonstrates reasonable conformance of cation exchange binding with HA when calcium affinity was quenched by phosphate. Overall, these data portray IgG retention on HA as embodying cation exchange retention roughly equivalent with dedicated cation exchangers, weak calcium affinity, very weak electrostatic repulsion between HA phosphate and IgG carboxyls, and no electrostatic interactions with HA calcium (in the presence of phosphate). Positive cooperativity between cation exchange and calcium affinity dominates retention. This interpretation, especially concerning cooperativity, is also consistent with published retention data in the form of dynamic binding capacity values. These data indicate much weaker calcium affinity than intact IgG, but comparison with retention on the cation exchanger demonstrates that cooperative enhancement by calcium affinity is still substantial. Even at 40 mM phosphate, sufficient calcium affinity remained to boost retention nearly threefold over the cation exchanger, to 17.2 mS. From 10 to 80 mM phosphate, the Fab retention curve essentially overlapped with the lysozyme curve (compare with Fig. 2 ), illustrating Fab’s dominant retention by cation exchange. The p I of Rituxan Fab was about 10, and its mass is only about 33% of intact IgG, both of which would be expected to reduce its retention compared with intact IgG, as shown. These observations are supported by the behavior of Fab in phosphate gradients at different chloride concentrations ( Table 2, Fig. 5 ). The early part of the Fab curve overlaps with lysozyme ( Fig. 3 ), illustrating cation exchange retention. With increasing sodium chloride concentrations, the curve becomes more parallel with the line indicating conductivity at gradient start.
The high conductivities at which antibodies elute in chloride gradients requires that the HA step be placed at the end of the purification process, or that the eluate be buffer exchanged to permit compatibility with a subsequent ion exchange step. The phosphate-chloride grid approach may also be applied in purification process development for fragmentary constructs. As an example from Fig. 4, Fab and Fc co-elute in a sodium chloride gradient at 10 mM phosphate, but are well separated in a chloride gradient at 20 mM phosphate where Fc fails to bind. This approach may also be useful in the purification of recombinant constructs from cell culture media, where BSA is a frequent contaminant. In the event that BSA co-elutes with a given product in a phosphate gradient absent sodium chloride, addition of chloride will cause the construct to elute earlier while leaving BSA retention essentially unaltered ( Table 2 ). For any application, the phosphate-chloride grid approach provides a systematic characterization tool that can be used to support process validation. With sodium chloride gradients for example, the variation in the vertical offset between two contaminants over a specified range of phosphate concentration illustrates the sensitivity of the separation to variations within that range. Grid results can thus be used to document the quantitative response of the process to any pertinent variable, and to document that process specifications lie within a range where routine variations in such variables will not adversely affect process control. Both approaches represent a rational data-driven approach to process design and validation, and both should lead to the same conclusions. Robotic systems produce more data points per unit time, but with substantial overhead costs. The grid approach can be employed at lower cost and offers the advantage of being able to model flow rate-dependent variables such as capacity and resolution.
Statistical design of experiments can be used to reduce the number of experiments for either approach, but results from the present study reveal an important caution: the effects of phosphate concentration are not linear over a broad range. A preliminary series of experiments should therefore be conducted to identify a narrow range of interest, within which design of experiments may be applied confidently. 4 Concluding remarks Results from this and other recent studies recommend several refinements to retention models that have been previously suggested for IgG. This documents that very weak calcium affinity interactions can be eluted exclusively by increasing conductivity, and suggests that stronger calcium affinity interactions are probably affected to some degree. Electrostatic repulsion between IgG carboxyls and HA phosphate occurs simultaneously with the dominant interactions of calcium affinity and phosphoryl cation exchange, but appears to be damped out at low conductivities and probably has a negligible effect on selectivity under typical elution conditions. All electrostatic interactions between protein charged residues and HA calcium are probably blocked by its strong complexation with mobile phase phosphate. While this model provides new insights concerning the interaction of IgG with HA, it remains far from complete and far from predictive. Many elements of HA interactivity with proteins remain inadequately characterized. Calcium affinity also requires more extensive characterization. Whether amino groups are also involved, including those in the peptide backbone, as in interactions between calcium and EDTA or EGTA, also remains to be determined. Hopefully, the substantial and growing economic importance of therapeutic antibodies, combined with the compelling abilities of HA for their purification, will encourage investigators to address these and other remaining questions. Capto MMC media proved capable of supporting adequate capture and significant dimer removal, although both loading and elution selectivity varied dramatically with the amount of supernatant applied to the column. An anion-exchange step was included to fortify overall virus and DNA removal. These results illustrate the value of multimodal chromatography methods when affinity chromatography methods are lacking and conventional alternatives prove inadequate. Examples include Fab, F(ab')2, single-chain (sc) Fv, bis-scFV, diabodies, minibodies, and single-domain antibodies ( 1 ).
Their small size potentially gives them access to tissues that are poorly accessible by intact antibodies; rapid clearance from blood and nontargeted tissues; lower immunogenic response; and eye-drop, inhalant, or oral administration. We report here on purification of an affinity-matured, humanized, antiprostate stem-cell antigen (PSCA) minibody for first-in-human clinical studies. This minibody and an earlier chimeric version have demonstrated excellent high-contrast microPET imaging for PSCA-positive human prostate, pancreatic, and bladder cancers in animal models ( Figure 1 ) ( 2 ). The overall structure is reminiscent of IgG, bivalent but only half the size. Although beneficial for effectiveness of the application, this architecture lacks the binding sites for protein A. Thus, the principal enabling tool for IgG platform purification is inapplicable, thereby presenting a greater challenge to process developers. Tumor locations are indicated by yellow arrows. Multimodal (mixed-mode) chromatography methods have existed since the 1950s ( 3 ). Hydroxyapatite (HA) is the archetype of multimodal chromatography methods, combining cation exchange and metal affinity. ( 4 ). It is also a prime example of the barriers that need to be overcome for mixed modes to become mainstream tools. HA's selectivity was recognized as unique from its introduction, but a lack of practical knowledge concerning its binding mechanisms long delayed the development of scouting pathways that fully revealed its abilities. That discouraged process developers who might have benefited from its capabilities. Successes in recombinant immunotherapy have stimulated introduction of other mixed-mode media.
Charged-hydrophobic mixed modes began to appear in the 1980s, mostly as hopeful protein A replacements. Such products arrive at a propitious moment: HA's successes have made process developers willing to confront the complexity of mixed modes and provided a conceptual framework for exploring multimodal interactions. High-throughput screening and statistical design of experiments (DoE) enable rapid accumulation of process-pertinent data ( 7, 25, 26 ). And the well-defined chemical structures of mixed-mode ligands provide valuable guidance on the types of eluting agents that promise useful results ( 27 ). Materials and Methods Cell Culture Anti-PSCA minibody (clone A11.1 2C3) was grown in NS0 cells using media supplemented with 2% fetal bovine serum. It is effective even at physiological pH and conductivity levels, at which its affinity for IgG and minibodies is nil. Used media can be discarded. We used 20 mM MES for buffering at pH 6, 20 mM Hepes at pH 7, and 20 mM Tris at pH 8. CX experiments (pH 4.5) were conducted in 20 mM sodium acetate. For an initial scouting run, the column was equilibrated to 10 mM sodium phosphate at pH 6.5, eluted in a linear gradient to 250 mM phosphate, and cleaned with 500 mM phosphate. A second run was conducted by equilibrating to 20 mM sodium phosphate at pH 6.5, eluting with a linear gradient to 20 mM phosphate and 1 M NaCl, then cleaning with 500 mM phosphate. Sample was prepared by 1:1 dilution of CCS with 50 mM MES at pH 6.0. The column was equilibrated with the same buffer, eluted with a linear gradient to 50 mM MES and 500 mM arginine, then cleaned with 2 M guanidine at pH 5.5. We used the higher MES concentration to compensate for buffer capacity of the MMC carboxyl group. Analysis We ran reduced and nonreduced sodium-dodecyl-sulfate polyacrylamide gel electrophoresis (SDS-PAGE) on precast Ready Gel 10% Tris-HCl Ready gels from Bio-Rad. It takes longer than you expect to install, if you want to install it right. Depending on your experience 30 minutes to an hour. The instructions call for the double pole 50 amp circuit breaker to be installed close as to the main breaker as possible. In our case we had to move two single pole 20 amp Breakers down which go to Kitchen GFI. And use those positions to install the 50 amp breaker next to the main breaker. Using Pliers broke out two filler tabs for the 20 amp breakers moved down. Instructions also calls for a liquid tight connection to box but there is no gasket included to make a liquid tight connection. If your breaker panel is indoors and there are no water pipes near the panel, you may be able to install it without a liquid tight connection. Or use a silicone RTV gasket sealant to seal the unit tight to the exterior of the panel box. This was the option we chose. And we mounted the unit at the bottom of the box to protect it from the rain and weather. Used a good sized bead so that the RTV squeezed out and into the inside of box completely sealing unit to box. Used hammer and the flat blade screwdriver to knock out the plug at the bottom nearest to the back and pliers to pull it off. Sometimes there is a burr that is left and it will need to be filed smooth. The wires supplied on the unit are about 18 inches long (45 cm) so we had to lengthen the red and black wires to reach to the 50 Amp Breaker at the top near the main breaker. Although the unit comes with 14 AWG stranded wires up sized to 12 AWG wire to reduce resistance. Do not use automotive grade 14 gauge stranded the insulation is not rated for AC voltage. We striped the wires and used yellow wire nuts that make the connections to the added wire. Then for additional safety wrapped electrical tape in the same direction we turned the nuts. Then fold it over the nut on to the wire and taped over for additional safety. Now house is fully protected against surges.

I installed it flush to my electrical panel using one of the free knock-outs. Installation took all of about 15 minutes from beginning to end. You will have to purchase a double-poled 50 amp breaker which was about 10.00. Wiring is very straight forward however a word of caution. Hopefully I will never have to find out if it works!Purchased a Siemens 50 amp double-pole breaker (you will need to purchase a breaker that is compatible with your own load center). Installation was very easy. Shut off power at the main breaker inside the load center. Flipped the 50 amp breaker off before installing. Installed 50 amp breaker onto bus bar. Mounted Eaton surge protector (how you mount it will vary based on how one’s load center is configured). Connected green wire to ground bar, white (neutral) wire to neutral bar, red wire to one of the 50-amp breakers, and the black wire to the other 50-amp breaker. Checked all wiring. Flipped the main breaker on. Flipped the 50 amp breaker on. Everything is good. LED lights on the surge protector are both on. As always, if you are not familiar with electricity and installing electrical components, please hire a certified electrician.I was unable to properly secure the 14 AWG wires to the 50 ampere circuit breaker because wire gauge was too small for the breaker terminal screws which are designed for 6 AWG wiring. I had to tin the two wires and make a solder loop to increase the size of the wires that would allow me to secure properly them to the 50 ampere circuit breaker terminals. Can a 14 AWG wire be installed using a 50 ampere circuit breaker? Yes. The connecting wires do not carry load current.

But it also provides protection till the refrigerator, microwave and air fry oven which all have electronic control boards and adding a hundred twenty volt surge protector is not easily done. The reason down grades to 4 stars is because the wire length is too short and had to add on to make it work in this application and they did not provide a gasket which are available from McMaster-Carr and are intended for a 1 inch hose washer. Wasn't about to buy 100 pieces for one installation and pay shipping. If you have everything on hand it will take you about 30 minutes to one hour to install it. But because we had to run back to the store to get additional wire it took us 2 hours. It's better to buy a little more wire and cut off the excess then to have to go to the store again. Saw the surge protector in action. A surge came from the Utility side (this is why you connected closest to the main) and the surge protector acted immediately and shunted the surge to ground. Lights did not dim appreciably but didn't notice our computers and TVs turn off just for a second then turn back on. The surge protector work the way it was supposed to. When it's shunted the surge to ground there was a temporary loss of power, but once the search had passed. The Protector returned back to normal mode.And my son's computer did a check to make sure everything was running okay. We also have standby power supplies with surge protection on our computers. That extra effort to install it correctly protected all our electronics and the new washer and dryer we just purchased.Easy to install ( I'm electrician) unless you’re an electrician, or have previous experience working with electric panels and know what the heck you’re doing, I highly recommend hiring an electrician for this job.
